Is inheritable genetic modification the new dividing line in gene therapy? The editors of this searching investigation, representing clinical medicine, public health and biomedical ethics, have established a distinguished team of scientists and scholars to address the issues from the perspectives of biological and social science, law and ethics, including an intriguing Foreword from Peter Singer. Their purpose is to consider how society might deal with the ethical concerns raised by inheritable genetic modification, and to re-examine prevailing views about whether (...) these procedures will ever be ethically and socially justifiable. The book also provides background to define the field, and discusses the biological and technological potential for inheritable genetic modification, its limitations, and its connection with gene therapy, cloning, and other reproductive interventions. Until recently, retention of introns in mature mRNAs has been regarded as a consequence of mis‐splicing. Intron‐retaining transcripts are thought to be non‐functional because they are readily degraded by nonsense‐mediated decay. However, recent advances in next‐generation sequencing technologies have enabled the detection of numerous transcripts that retain introns. As we review herein, intron‐retaining mRNAs play an essential conserved role in normal physiology and an emergent role in diverse diseases. Intron retention should no longer be overlooked as a key mechanism that (...) independently reduces gene expression in normal biology. This volume is a collection of essays by various contributors in honor of the late Laurence Berns, Richard Hammond Elliot Tutor Emeritus at St. John's College, Annapolis. The essays address the literary, political, theological, and philosophical themes of his life's work as a scholar, teacher, and constant companion of the "great books.".

Continuous recordings of brain electrical activity were obtained from a group of 176 patients throughout surgical procedures using general anesthesia. Artifact-free data from the 19 electrodes of the International 10/20 System were subjected to quantitative analysis of the electroencephalogram (QEEG). Induction was variously accomplished with etomidate, propofol or thiopental. Anesthesia was maintained throughout the procedures by isoflurane, desflurane or sevoflurane (N = 68), total intravenous anesthesia using propofol (N = 49), or nitrous oxide plus narcotics (N = 59). A set (...) of QEEG measures were found which reversibly displayed high heterogeneity of variance between four states as follows: (1) during induction; (2) just after loss of consciousness (LOC); (3) just before return of consciousness (ROC); (4) just after ROC. Homogeneity of variance across all agents within states was found. Topographic statistical probability images were compared between states. At LOC, power increased in all frequency bands in the power spectrum with the exception of a decrease in gamma activity, and there was a marked anteriorization of power. Additionally, a significant change occurred in hemispheric relationships, with prefrontal and frontal regions of each hemisphere becoming more closely coupled, and anterior and posterior regions on each hemisphere, as well as homologous regions between the two hemispheres, uncoupling. All of these changes reversed upon ROC. Variable resolution electromagnetic tomography (VARETA) was performed to localize salient features of power anteriorization in three dimensions. A common set of neuroanatomical regions appeared to be the locus of the most probable generators of the observed EEG changes. (shrink)

The present research seeks to better understand research conditions in laboratory research, with special attention paid to the informed consent process and experimenter characteristics. The first study tested the impact of language perspective and experimenter demeanor upon participant retention of the informed consent information, attitudes toward the research project, and performance on experimental tasks. The second study examined the impact of experimenter attire. Across the two studies, our results suggest that there was no impact of language perspective, whereas the number (...) of other participants in the laboratory, experimenter attire, and experimenter demeanor influence participant behaviors in the laboratory. Each essay is a comprehensive introduction to the thought of a noted American on the fundamental meaning of the American regime.

We report an experiment investigating the “special-process” theory of insight problem solving, which claims that insight arises from non-conscious, non-reportable processes that enable problem re-structuring. We predicted that reducing opportunities for speech-based processing during insight problem solving should permit special processes to function more effectively and gain conscious awareness, thereby facilitating insight. We distracted speech-based processing by using either articulatory suppression or irrelevant speech, with findings for these conditions supporting the predicted insight facilitation effect relative to silent working or thinking (...) aloud. The latter condition was included to investigate the currently contested effect of “verbal overshadowing” on insight, whereby thinking aloud is claimed to hinder the operation of special, non-reportable processes. Whilst verbal overshadowing was not evident in final solution rates, there was nevertheless support for verbal overshadowing up to and beyond.. (shrink)

Regulation of the cytoplasmic stability of mRNAs has recetly been identified as a major control mechanism which governs mRNA levels in a variety of eukaryotic systems. In this review we discuss what is known about several experimental systems that exhibit regulated mRNA stability, describe the mechanisms that cells may use to achieve control of mRNA degradation, and suggest areas of future investigation likely to provide new insights into this process.

Humans, including preschool children, exhibit role-based relational reasoning, of which analogical reasoning is a canonical example. The connectionist model proposed in the target article is only capable of conditional paired-associate learning.

People consistently act in ways that harm the environment, even when believing their actions are environmentally friendly. A case in point is a biased judgment termed the negative footprint illusion, which arises when people believe that the addition of “eco-friendly” items to conventional items, reduces the total carbon footprint of the whole item-set, whereas the carbon footprint is, in fact, increased because eco-friendly items still contribute to the overall carbon footprint. Previous research suggests this illusion is the manifestation of an (...) “averaging-bias.” We present two studies that explore whether people’s susceptibility to the negative footprint illusion is associated with individual differences in: environment-specific reasoning dispositions measured in terms of compensatory green beliefs and environmental concerns; or general analytic reasoning dispositions measured in terms of actively open-minded thinking, avoidance of impulsivity and reflective reasoning. A negative footprint illusion was demonstrated when participants rated the carbon footprint of conventional buildings combined with eco-friendly buildings and conventional cars combined with eco-friendly cars. However, the illusion was not identified in participants’ ratings of the carbon footprint of apples. In Studies 1 and 2, environment-specific dispositions were found to be unrelated to the negative footprint illusion. Regarding reflective thinking dispositions, reduced susceptibility to the negative footprint illusion was only associated with actively open-minded thinking measured on a 7-item scale and 17-item scale. Our findings provide partial support for the existence of a negative footprint illusion and reveal a role of individual variation in reflective reasoning dispositions in accounting for a limited element of differential susceptibility to this illusion. This chapter focuses on John Locke's relation to scholasticism. It explores who the schoolmen referred to by Locke were, and what he might have learned from them, particularly with respect to topics in metaphysics, logic, and language. The chapter considers the Oxford curriculum which provided the framework for Locke's years of study and teaching there, as there is little reason to believe that he enriched his acquaintance with the schoolmen in his later career. The topic of substance was raised (...) both in metaphysics textbooks and in logic textbooks, but the material in both was largely similar. The chapter considers some of Locke's remarks and focuses on what the scholastics had to say. Locke goes on to claim that the very use of syllogistic depends on the prior ability to see the connections between ideas. The area in which Locke showed the most obvious influence of scholasticism is that of language. (shrink)

The ease with which genotyping technologies generate tremendous amounts of data on research participants has been well chronicled, a feat that continues to become both faster and cheaper to perform. In parallel to these advances come additional ethical considerations and debates, one of which centers on providing individual research results and incidental findings back to research participants taking part in genetic research efforts. In 2006 the Industry Pharmacogenomics Working Group offered some ‘Points-to-Consider’ on this topic within the context of the (...) drug development process from those who are affiliated to pharmaceutical companies. Today many of these points remain applicable to the discussion but will be expanded upon in this updated viewpoint from the I-PWG. The exploratory nature of pharmacogenomic work in the pharmaceutical industry is discussed to provide context for why these results typically are not best suited for return. Operational challenges unique to this industry which cause barriers to returning this information are also explained. (shrink)

He insists that our cognition is rooted in sense-perception and that [End Page 673] it is normally reliable. He (...) insists that our moral life is grounded in our nature both as physical beings, able to feel pain and pleasure, and as social beings, depending on each other for our welfare. But he is also strongly supernaturalist. Human nature by virtue of its cognitive orientation toward the physical is not able through its own powers to know God. Still less is human nature able to achieve moral virtue by its own powers. This is not just because we are cut off from God by original sin; rather, there is a primal lack that was there even before the Fall. To achieve our true end, both cognitive and moral, we depend on God’s free gift of grace which elevates our nature and renders it able both to know God through the beatific vision in Heaven and to respond to God through loving, virtuous action. The gift of faith becomes central to human nature and destiny in a way that was to worry later thinkers such as Duns Scotus, who pertinently asked how our intellect can have the ultimate end of knowing God if it has to be made a different intellect before it achieves that end.Jenkins does not worry about the point raised by Scotus. Instead, he sets out to give an interpretation of the Summa theologiae which will help us understand more deeply the interplay between the natural and the supernatural in Aquinas. He begins by taking advantage of new readings of Aristode’s Posterior Analytics which present Aristode’s account of the demonstration which moves from cause to effect, not just as a pedagogic device for presenting material obtained by moving from effect to cause, but as a recipe for cognitive restructuring. Certainly we have to begin with mere observation, and with the piecemeal discovery of causes, but to achieve true scıentia (to use the medieval Latin term for organized, necessary knowledge) we need to acquire a new vision of how reality is structured and to see how effects flow necessarily from their causes. True demonstration is an expression of this new vision. Jenkins applies this reading of Aristotle to Aquinas. He takes with absolute seriousness Aquinas’s claim at the beginning of the Summa theologıae that theology too is an Aristotelian scientıa, and he sees the work as presenting a recipe for the cognitive restructuring of apprentice theologians (those to whom the work is addressed).The scientıa of the Summa theologiae differs from the scientia of the Posterior Analytics in two ways. First, Aquinas allows the term scientia to have an extended sense, so that God too can have scientia, even though he does not move from principles to conclusions by a process of syllogistic demonstration, but instead grasps all that is knowable in one simple act. Second, the basic principles which are the foundation of theological demonstration are the articles of faith, especially those of the creeds, which cannot be known by us in any naturalistic way. It is through God’s infused light that we assent to these articles as divinely revealed. Drawing on a landscape analysis of existing data-sharing initiatives, in-depth interviews with expert stakeholders, and public deliberations with community advisory panels across the U.S., we describe features of the evolving medical information commons. We identify participant-centricity and trustworthiness as the most important features of an MIC and discuss the implications for those seeking to create a sustainable, useful, and widely available collection of linked resources for research and other purposes.

Theories of analogical reasoning have viewed relational structure as the dominant determinant of analogical mapping and inference, while assigning lesser importance to similarity between individual objects. An experiment is reported in which these two sources of constraints on analogy are placed in competition under conditions of high relational complexity. Results demonstrate equal importance for relational structure and object similarity, both in analogical mapping and in inference generation. The human data were successfully simulated using a computational analogy model (LISA) that treats (...) both relational correspondences and object similarity as soft constraints that operate within a limited-capacity working memory; but not with a model (SME) that treats relational structure as pre-eminent. Salespeople have long been considered unique employees. They tend to work apart from each other and experience little daily contact with supervisors and other organizational employees. Additionally, salespeople interact with customers in an increasingly complex and multifunctional environment. This provides numerous opportunities for unethical behavior which has been chronicled in the popular press as well as academic research. Much of the research in sales ethics has relied on conceptual foundations which focus on individual and organizational influencers on ethical decision making. (...) While significant, contributors to this research suggest that alternative theoretical perspectives and methods of investigation should be utilized and call for more research on the status of professional selling as a whole. We answer this call by exploring an alternative and complementary perspective based on the theory of occupational choice, social learning, and work groups to gain insight on how the sales profession evolves as its own subculture that extends beyond individual and organizational boundaries. First, we discuss the characteristics of the sales profession and empirically examine the relationship between typical individual and organizational factors and sales professionals’ perceptions of ethical behavior. Second, we offer a theoretical explanation that our findings may be due to how salespeople choose and are socialized into the subculture of the sales profession. Third, we examine this theoretical perspective via qualitative in-depth interviews with experienced sales professionals. Results and implications are discussed in terms of a sales profession code of ethics and future research directions. (shrink)

John Locke was one of the towering philosophers of the Enlightenment and arguably the greatest English philosopher. Many assumptions we now take for granted, about liberty, knowledge and government, come from Locke and his most influential works, _An Essay Concerning Human Understanding_ and _Two Treatises of Government_. In this superb introduction to Locke's thought, E.J. Lowe covers all the major aspects of his philosophy. Whilst sensitive to the seventeenth-century background to Locke's thought, he concentrates on introducing and assessing Locke (...) in a contemporary philosophical setting, explaining why he is so important today. Beginning with a helpful overview of Locke's life and times, he explains how Locke challenged the idea that the human mind and knowledge of the external world rested on innate principles, laying the philosophical foundations of empiricism later taken up by Berkeley and Hume. Subsequent chapters introduce and critically assess topics fundamental to understanding Locke: his theories of substance and identity, language and meaning, philosophy of action and free will, and political freedom and toleration. In doing so, he explains some of the more complex yet pivotal aspects of Locke's thought, such as his theory that language rests on ideas and how Locke's theory of personal identity paved the way for modern empirical psychology. A final chapter assesses Locke's legacy, and the book includes a helpful chronology of Locke's life and glossary of unfamiliar terms. (shrink)

‘Water is H2O’ is one of the most frequently cited sentences in analytic philosophy, thanks to the seminal work of Saul Kripke and Hilary Putnam in the 1970s on the semantics of natural kind terms. Both of these philosophers owe an intellectual debt to the empiricist metaphysics of John Locke's Essay Concerning Human Understanding, while disagreeing profoundly with Locke about the reality of natural kinds. Locke employs an intriguing example involving water to support his view that kinds (or ‘species’), (...) such as water and gold, are the workmanship of the human mind. This is the point of his story about a winter visitor to England from Jamaica, who is astonished to find that the water in his basin has turned solid overnight, and proceeds to call it ‘hardened water’. Locke criticizes this judgement, maintaining that it is more consonant with common sense to regard water and ice as different kinds of substance. Putnam, by implication, disagrees. Deploying his imaginary example of Twin Earth—a distant planet where a watery-looking substance, XYZ, rather than H2O, fills the oceans and rivers—he maintains that common sense supports the judgement that XYZ and H2O, despite their superficial similarity, are not the same kind of substance, precisely because their molecular compositions are different. Here it will be argued that both views are mistaken, but that, in this dispute, Locke has more right on his side than his modern opponents do. (shrink)
